Eosin Differentiation

In the H&E stain, eosin staining is commonly followed by two changes of 95% ethanol. The ethanol+water combination aids with removing excess eosin from the tissue, thus acting as a differentating step. It is important to ensure dehydration is complete after these steps as excess water in xylene can continue the eosin differentiation process. This can be seen after coverslipping as a pink haze on the slide.
